Despite more than $30 billion in research funding since 1984, no cure, prevention, or effective long-term treatment exists.<\/p>\r\n\r\n<p style=\"margin-bottom: 40px;\">It\u2019s no secret that Alzheimer\u2019s research hasn\u2019t progressed as quickly as we\u2014and the many families affected\u2014would like. That\u2019s in part due to tunnel vision.<\/p>\r\n\r\n<div class=\"text-wrap-container\">\r\n<div class=\"pull-right-wrap\">\r\n<figure><img decoding=\"async\" style=\"width: 100%;\" src=\"\/wp-content\/uploads\/2025\/09\/25-alzheimers-plaque-research.jpg\" \/><\/figure>\r\n<p style=\"font-size: .9em; line-height: 1.3; padding: 10px;\">Most Alzheimer\u2019s research has focused on the plaques (white) and tangles (dark blue) that can form in and around neurons (light blue) at later stages of the disease.<\/p>\r\n\r\n<\/div>\r\n<p><strong>Focus almost exclusively on neurons.<\/strong> Most research on brain function and cognitive decline is focused exclusively on neurons. Yet, in reality, neurons make up only half of the brain\u2019s total cell count. The other half are glial cells, which support and protect neurons and undoubtedly play an underappreciated role in maintaining brain health.<\/p>\r\n\r\n<p><strong>Overemphasis on amyloid plaques and tau tangles.<\/strong> These abnormal clumps of proteins in the brain were first observed by Alois Alzheimer himself in 1906 and thus became the defining biomarkers of the disease.<\/p>\r\n\r\n<p>For decades, these proteins were the focus of nearly all Alzheimer\u2019s research and more than 400 failed clinical trials.<\/p>\r\n\r\n<p>There are two recently approved monoclonal antibody therapies that reduce amyloid plaques, but the benefit to patients is modest at best.<\/p>\r\n\r\n<p><strong>More attention given to rarer form of Alzheimer\u2019s.<\/strong> Most Alzheimer\u2019s research has focused on the familial type, which is a rare, early-onset form caused by inherited gene mutations. Yet more than 90% of cases are sporadic, not inherited.<\/p>\r\n\r\n<p><em>The need for a transformative approach has never been more urgent.<\/em><\/p>\r\n\r\n<\/div>\r\n<!-- end challenge -->\r\n\r\n<hr \/>\r\n\r\n<p style=\"color: #653ac8; font-size: 20px; text-transform: uppercase;\"><strong>The Salk Institute is rewriting the narrative of Alzheimer\u2019s disease research<\/strong><\/p>\r\n\r\n<div class=\"text-wrap-container\">\r\n<div class=\"pull-right-wrap\">\r\n<figure><img decoding=\"async\" style=\"width: 100%;\" src=\"\/wp-content\/uploads\/2025\/09\/25-alzheimers-salk-approach.jpg\" \/><\/figure>\r\n<p style=\"font-size: .9em; line-height: 1.3; padding: 10px;\">Salk\u2019s approach pulls back the lens to consider other potential drivers of disease, including astrocytes (purple), microglia (cyan), immune cells (orange), inflammation (red), and more.<\/p>\r\n\r\n<\/div>\r\n<p><strong>Chronic inflammation: A hidden threat in the brain.<\/strong> Chronic inflammation is a sustained immune response in the brain over an individual\u2019s lifetime, and that long-term damage can ultimately manifest as Alzheimer\u2019s disease. The Salk team also identified two key factors as key contributors to this harmful inflammation: genome instability and dysregulated energy metabolism.<\/p>\r\n\r\n<p>Salk scientists believe that inflammation, genome stability, and energy metabolism are directly connected and influence one another.<\/p>\r\n\r\n<p>Even at baseline, human brains require enormous amounts of energy\u2014approximately 20% of the body\u2019s total energy consumption, about twice that of other primates. Our brains are already running in energetic hyperdrive. So, when life throws an added stress on top of that, such as the need for extra DNA repair, or as mitochondria naturally decline with age, inflammation increases. Ultimately, this stress makes the brain more susceptible to Alzheimer\u2019s.<\/p>\r\n\r\n<p>Salk\u2019s approach\u2014widening the research lens to focus on chronic inflammation throughout the brain\u2014will allow researchers to identify biomarkers for early signs of the disease and new opportunities for intervention with next-generation therapeutics.<\/p>\r\n\r\n<\/div>\r\n<!-- what drives alz -->\r\n<div class=\"row\" style=\"background-color: #e5ebfb; margin: 40px 0; padding: 20px;\">\r\n<h2 style=\"color: #653ac8;\">What drives Alzheimer\u2019s disease?<\/h2>\r\n<!-- bullet 1 -->\r\n<div style=\"margin: 20px 0;\">\r\n<div style=\"float: left; width: 3%; margin-right: 6px;\"><img decoding=\"async\" style=\"width: 100%;\" src=\"\/wp-content\/uploads\/2025\/09\/25-YoA-circle-arrow-turquoise.png\" \/><\/div>\r\n<div style=\"float: left; width: 95%;\">\r\n<p style=\"line-height: 1.4;\"><em>Chronic inflammation.<\/em> The body\u2019s natural defense response to injury or infection can cause harm if it persists as chronic inflammation or is inappropriately directed.<\/p>\r\n\r\n<\/div>\r\n<div style=\"clear: both;\"><\/div>\r\n<\/div>\r\n<!-- end bullet 1 -->\r\n<p style=\"margin-bottom: 0;\"><strong>What contributes to harmful chronic inflammation in the brain?<\/strong><\/p>\r\n<!-- bullet 2 -->\r\n<div style=\"margin: 10px 0 20px 30px;\">\r\n<div style=\"float: left; width: 3%; margin-right: 6px;\"><img decoding=\"async\" style=\"width: 100%;\" src=\"\/wp-content\/uploads\/2025\/09\/25-YoA-circle-arrow-pink.png\" \/><\/div>\r\n<div style=\"float: left; width: 90%;\">\r\n<p style=\"line-height: 1.4;\"><em>Genome instability.<\/em> When genomes and chromosomes are unstable, errors can occur in the way our genetic information (DNA) is copied and passed on.<\/p>\r\n\r\n<\/div>\r\n<div style=\"clear: both;\"><\/div>\r\n<\/div>\r\n<!-- end bullet 2 -->\r\n\r\n<!-- bullet 3 -->\r\n<div style=\"margin: 20px 0 20px 30px;\">\r\n<div style=\"float: left; width: 3%; margin-right: 6px;\"><img decoding=\"async\" style=\"width: 100%;\" src=\"\/wp-content\/uploads\/2025\/09\/25-YoA-circle-arrow-pink.png\" \/><\/div>\r\n<div style=\"float: left; width: 90%;\">\r\n<p style=\"line-height: 1.4;\"><em>Dysregulated energy metabolism.<\/em> If not properly regulated, the process by which our cells use mitochondria to generate energy malfunctions, leaving cells depleted and unable to maintain healthy functions.<\/p>\r\n\r\n<\/div>\r\n<div style=\"clear: both;\"><\/div>\r\n<\/div>\r\n<!-- end bullet 3 -->\r\n\r\n<\/div>\r\n<!-- end what drives alz -->\r\n\r\n<p style=\"color: #653ac8; font-size: 20px; text-transform: uppercase;\"><strong>Cross-disciplinary collaboration drives innovation<\/strong><\/p>\r\n\r\n<p>Here are just a few examples of Salk studies that demonstrate our expertise in neuroscience, immunobiology, genomics, and metabolism, as well as our strengths in thinking creatively and collaboratively to find what few others are even looking for.<\/p>\r\n\r\n<p><strong>Aging biology and metabolism experts<\/strong> discovered that when telomeres (the end caps of chromosomes) become very short with age, they communicate with mitochondria. This communication triggers a complex set of signaling pathways and initiates an inflammatory response. The team is now determining how this crosstalk between genomes, mitochondria, and inflammation plays a role in Alzheimer\u2019s disease.<\/p>\r\n\r\n<p><strong>Neuroscientists<\/strong> found that energy-intense neuron DNA repairs are not random but instead focus on protecting certain genetic \u201chot spots\u201d that appear to play a critical role in neural identity and function. As cellular energy production declines as we age, neurons struggle to repair their genomes. Accordingly, the team is now unraveling the interplay between genomes and energy metabolism to determine its role in Alzheimer\u2019s disease.<\/p>\r\n\r\n<p>Salk neuroscientists have also discovered that astrocytes, the most common type of glial cell, are crucial for shaping communication across the brain. They also found that in Alzheimer\u2019s patients, astrocytes are less able to create or strengthen synapses, ultimately disrupting brain communication. The scientists are now testing whether boosting astrocyte function in the Alzheimer\u2019s brain can restore synaptic function and delay disease progression.<\/p>\r\n\r\n<p><strong>Immunologists<\/strong> are investigating how lifelong exposure to infections and other foreign agents influences immune cell infiltration and brain inflammation, potentially contributing to the development of Alzheimer\u2019s disease.<\/p>\r\n\r\n<p>Salk scientists have revealed that the body has evolved mechanisms to tolerate infections by maintaining physiological function and repairing tissue damage\u2014both of which are necessary for survival. Beyond infections, this approach reshapes our perception of health\u2014not merely as the absence of disease but as an active, mechanistic process of maintaining resilience and endurance.
