En un estudio publicado en un pr\u00f3ximo n\u00famero de Nature Genetics, el equipo de Salk, dirigido por <a href=\"\/es\/faculty\/belmonte.html\/\">Juan Carlos Izpis\u00faa Belmonte<\/a>, Ph.D., profesora en el Laboratorio de Expresi\u00f3n G\u00e9nica, identific\u00f3 un nuevo factor que vincula las se\u00f1ales del desarrollo temprano con la funci\u00f3n de los cilios y su papel en el control de la especificaci\u00f3n izquierda-derecha en peces cebra.<\/p>\n<p>\u201cCuando alteramos la funci\u00f3n del gen *duboraya*, observamos problemas en la formaci\u00f3n de cilios, aunque el producto g\u00e9nico en s\u00ed no forma parte de la estructura. Esto abre un nuevo campo de investigaci\u00f3n\u201d, dice Belmonte.<\/p>\n<p>Las cilias han sido conocidas por los bi\u00f3logos celulares durante m\u00e1s de cien a\u00f1os. Belmonte est\u00e1 convencido de que estas humildes estructuras, que hasta hace poco han sido ignoradas tanto por fisi\u00f3logos como por bi\u00f3logos moleculares, est\u00e1n listas para ocupar un lugar central en el campo de la biolog\u00eda. Belmonte explica: \u201cCuando se perjudica la funci\u00f3n de las cilias o el flujo de las cilias, se crean problemas sustanciales en todo el cuerpo\u201d.\u201d <\/p>\n<p>Estas simples estructuras en forma de l\u00e1tigo no solo participan de manera cr\u00edtica en la especificaci\u00f3n de la lateralidad izquierda-derecha durante el desarrollo, sino que ayudan a mover fluidos y mucosidad alrededor del cerebro, los pulmones, los ojos y los ri\u00f1ones, y son necesarias para el olfato, la vista y la reproducci\u00f3n. Enfermedades m\u00e9dicas, como la diabetes y la obesidad, se han relacionado con defectos estructurales en la arquitectura o en la funci\u00f3n de las cilias. Adem\u00e1s, evidencia reciente indica que las cilias pueden tener roles adicionales en el control del desarrollo esquel\u00e9tico y la funci\u00f3n cerebral.<\/p>\n<p>Cilios en la superficie exterior de la parte inferior del embri\u00f3n, un \u00e1rea llamada n\u00f3dulo ventral en mam\u00edferos, exhiben un movimiento caracter\u00edstico de giro que transporta mensajeros qu\u00edmicos hacia el lado izquierdo. Esto crea un gradiente de concentraci\u00f3n qu\u00edmica que indica a las c\u00e9lulas madre c\u00f3mo y d\u00f3nde desarrollarse. Cuando la funci\u00f3n de los cilios se ve afectada, \u00f3rganos como el coraz\u00f3n, los pulmones y el h\u00edgado pueden terminar en el lado equivocado del cuerpo. <\/p>\n<p>Cuando el investigador postdoctoral y autor principal Isao Oishi, Ph.D., busc\u00f3 genes en el pez cebra involucrados en el patr\u00f3n de izquierda-derecha de embriones tempranos, esperaba encontrar genes que codificaran componentes de cilios. \u201cEn cambio, encontramos un gen de cilio no estructural que influye en la funci\u00f3n de los cilios y que, entre otras cosas, caus\u00f3 problemas con el patr\u00f3n de izquierda\/derecha\u201d, dice. Nombr\u00f3 al gen duboraya en honor a la forma de la linterna japonesa duboraya, que adoptan los peces con una versi\u00f3n inactivada del gen a medida que se desarrollan. <\/p>\n<p>Oishi descubri\u00f3 que la duboraya es necesaria para la formaci\u00f3n de cilios completamente funcionales en la ves\u00edcula de Kupffer, el equivalente en peces del nodo ventral de los mam\u00edferos. Sin duboraya, los cilios se reduc\u00edan a tocones cortos, incapaces de crear el flujo en sentido contrario a las agujas del reloj necesario para establecer la izquierda frente a la derecha. La prote\u00edna duboraya, encontr\u00f3, es activada por la frizzled-2, un miembro de la v\u00eda de se\u00f1alizaci\u00f3n Wnt, altamente conservada, que orquesta las actividades de un gran n\u00famero de c\u00e9lulas durante el desarrollo embrionario. <\/p>\n<p>Belmonte explica: \u201cPudimos demostrar que genes que detectan su entorno externo o interno se comunican con genes estructurales que son responsables de fabricar los cilios y les dicen que batan de una u otra manera.
